SSC (Northern Blot, 20x) Preparation and Recipe
SSC (saline sodium citrate) buffer is commonly used in Northern blotting as a hybridization or wash buffer.
To prepare L of SSC (Northern Blot, 20x):

Sodium chloride (mw: 58.44 g/mol) | 175.3 g | 3 M |
Trisodium citrate (mw: 258.07 g/mol) | 77.4 g | 0.3 M |
- Prepare 800 mL of distilled water in a suitable container.
- Add 175.3 g of Sodium chloride to the solution.
- Add 77.4 g of Trisodium citrate to the solution.
- Add distilled water until the volume is 1 L.
- Adjust the pH to 7.0 with NaOH or HCl. Autoclave the solution and store it at room temperature. It will last for 1 yr or longer.
