Acetate Buffer (0.1 M, pH 5.0) Preparation and Recipe
Sodium acetate buffers are used for purification and precipitation of nucleic acids, as well as for protein crystallization and staining gels used in protein electrophoresis.
To prepare L of Acetate Buffer (0.1 M, pH 5.0):

Sodium Acetate (mw: 82.03 g/mol) | 5.772 g | 0.07 M |
Acetic Acid (mw: 60.05 g/mol) | 1.778 g | 0.03 M |
- Prepare 800 mL of distilled water in a suitable container.
- Add 5.772 g of Sodium Acetate to the solution.
- Add 1.778 g of Acetic Acid to the solution.
- Adjust solution to desired pH using 10N HCl (typically pH ≈ 5.0).
- Add distilled water until the volume is 1 L.
