Maleic Acid (1 M, pH 7.5) Preparation and Recipe

Maleic acid buffer is commonly used for Southern blot preparation and as a washing buffer in numerous applications involving nucleic acids. It has a shelf life of 6 months to a year.

To prepare  L of Maleic Acid (1 M, pH 7.5):
Change the value in the textbox above to scale the recipe volume

Table 1. Required components
ComponentAmountConcentration
Maleic Acid (mw: 116.07 g/mol)116.1 g1 M
Sodium chloride (mw: 58.44 g/mol)87.6 g1.5 M
Sodium Hydroxide (mw: 40.00 g/mol)40 g1 M
  1. Prepare 800 mL of dH2O in a suitable container.
  2. Add 116.1 g of Maleic Acid to the solution.
  3. Add 87.6 g of Sodium chloride to the solution.
  4. Add 40 g of Sodium Hydroxide to the solution.
  5. Add dH2O until the volume is 1 L.
  6. Sterilize by autoclaving at 15 psi for 20 minutes or by filtration. Store at room temperature. For washing applications, add 0.2% Tween-20 if desired.
