Maleic Acid (1 M, pH 7.5) Preparation and Recipe
Maleic acid buffer is commonly used for Southern blot preparation and as a washing buffer in numerous applications involving nucleic acids. It has a shelf life of 6 months to a year.
To prepare L of Maleic Acid (1 M, pH 7.5):

Sodium chloride (mw: 58.44 g/mol) | 87.6 g | 1.5 M |
Sodium Hydroxide (mw: 40.00 g/mol) | 40 g | 1 M |
- Prepare 800 mL of dH2O in a suitable container.
- Add 116.1 g of Maleic Acid to the solution.
- Add 87.6 g of Sodium chloride to the solution.
- Add 40 g of Sodium Hydroxide to the solution.
- Add dH2O until the volume is 1 L.
- Sterilize by autoclaving at 15 psi for 20 minutes or by filtration. Store at room temperature. For washing applications, add 0.2% Tween-20 if desired.
