MOPS-EGTA buffer has a variety of uses including membrane solubilization and fixation. MOPS is widely used in biology and biochemistry applications due to its stability, while EGTA is a chelating agent.

To prepare  L of MOPS-EGTA Buffer:
Change the value in the textbox above to scale the recipe volume

Table 1. Required components
ComponentAmountConcentration
MOPS (mw: 209.26 g/mol)20.9 g0.1 M
Magnesium sulfate (mw: 120.37 g/mol)0.24 g0.002 M
EGTA (mw: 380.35 g/mol)0.38 g0.001 M
Sodium chloride (mw: 58.44 g/mol)29.2 g0.5 M
  1. Prepare 800 mL of distilled water in a suitable container.
  2. Add 20.9 g of MOPS to the solution.
  3. Add 0.24 g of Magnesium sulfate to the solution.
  4. Add 0.38 g of EGTA to the solution.
  5. Add 29.2 g of Sodium chloride to the solution.
  6. Adjust the pH to 7.5 with NaOH.
  7. Add distilled water until the volume is 1 L.
  8. Sterilize the buffer using a 0.22-µm filter. The buffer can be stored at room temperature for several months.
