Helixyte Green (SYBR Green I)

20X Aqueous PCR Solution
SYBR® Green I (Thermo Fisher Scientific) and Helixyte™ Green are chemically equivalent, nucleic acid stains, with an excitation peak at 498 nm and an emission peak at 522 nm. The dye is useful for the detection of double-stranded DNA (dsDNA) in gel electrophoresis and quantitative PCR (qPCR) applications.

Staining mechanism and principle

Helixyte™ Green is an intercalating nucleic acid dye belonging to the cyanine dye family. It is fluorogenic, becoming highly fluorescent upon binding to the minor groove of dsDNA and, to a lesser extent, when bound to single stranded DNA (ssDNA) and RNA. It can be used to stain dsDNA in polyacrylamide and agarose gels in electrophoresis applications. It is also useful for monitoring qPCR, wherein fluorescence measured during the annealing or extension stage is proportional to the amount of PCR product. In these applications, often the dye is provided alongside the master mix.

Excitation / Emission

Helixyte Green and SYBR Green I have the same fluorescence spectrum, with an excitation peak at 498 nm and an emission peak at 522 nm. They are well excited by the 488 nm laser line and can be detected in the FITC channel.

Helixyte/SYBR Green versus Ethidium Bromide (EtBr)

Helixyte™ Green and SYBR Green stains are in principle safer and less mutagenic than EtBr because they are less cell permeable. Additionally, these dyes are experimentally more convenient due to having a green excitation wavelength, which is compatible with common lab instruments, such as the FITC channel on a fluorescence microscope or the 488 nm laser on a flow cytometer. This is in contrast to EtBr, which requires the more dangerous UV excitation wavelength.

SYBR Green I versus SYBR Green II

SYBR Green I has a strong affinity for dsDNA, whereas SYBR Green II has a strong affinity for RNA.
