The principle of Restriction Fragment Length Polymorphism (RFLP) involves observing variations in the lengths of DNA fragments produced when DNA samples are digested with specific restriction enzymes. If two organisms have differences in the distance between recognition sites of a particular enzyme, the resulting fragments will vary in length. By comparing these fragment patterns, similarities and differences can be identified, enabling differentiation between strains or species. RFLP markers work by distinguishing genomic DNA based on the presence or absence of specific sites where restriction enzymes cleave the DNA.
