What is the advantage of using a Southern blot over other methods?

Southern blotting doesn't depend on PCR to amplify DNA, allowing for the accurate sizing of repeat expansions that are too large for PCR amplification. Southern blot also has the ability to provide quantitative results, displaying both digested and undigested DNA molecules. Southern blot is also used to analyze multiple similar sequences in the genome.
