What factors should I consider when choosing reagents for live-cell imaging?

Posted September 11, 2024

Answer

Reagents can include fluorescent proteins that target specific cell components or small dyes that pass through cell membranes. The choice depends on whether one needs to visualize specific proteins or general cellular characteristics. Another factor are imaging parameters, as the time window and frequency of imaging should be optimized to get the best results while avoiding excessive damage to the cells. A third factor is duration of imaging. Some reagents are designed for experiments that last several hours or days, while others are meant for quick, end-point assays where cells are imaged right after staining. Lastly, one should consider the staining conditions. They must carefully determine the concentration of the staining reagent and the incubation time to balance effective staining with minimal cytotoxicity. This helps to preserve the overall health and functioning of cells.
