What factors should be considered when choosing the membrane for western blotting?

Answer

When choosing a membrane, one should consider that different membranes may exhibit different binding efficiencies for proteins of varying sizes. For example, PVDF membranes are suited for high MW proteins, nylon membranes are suited for nucleic acids, and nitrocellulose membranes are suited for middle to low MW proteins. One should also consider the transfer efficiency of the membrane to ensure that proteins are transferred effectively from the gel to the membrane. Additionally, if the experiment requires the need for stripping and reprobing the membrane for multiple targets, choose a membrane that is compatible (e.g. PVDF).
