What are the limitations of tank transfer technique?

One limitation is that the time required for wet tank transfer is typically at least one hour, although this can vary based on factors such as percent acrylamide, gel dimensions, target protein molecular weight, buffer conditions, and membrane characteristics. Alternatively, tank transfer can be run overnight at lower power settings. Another limitation is the use of a transfer buffer containing methanol can lead to an increase in hazardous lab waste due to its toxicity. Additionally, maintaining a cold temperature in the transfer tank apparatus is necessary to prevent sample overheating. This requires either space at 4°C or an apparatus such as a cooling coil with a refrigerated water recirculatory system.
