What are the limitations of Coomassie blue stains?

One disadvantage is the staining intensity can vary depending on the amino acid composition of the proteins, which makes it challenging to use Coomassie for accurately comparing the quantities of different proteins. Traditional Coomassie staining methods usually take a long time for staining and destaining to get a clear background. However, extended destaining can also diminish the intensity of the protein bands, resulting in reduced sensitivity. Another drawback of traditional Coomassie staining is its low reproducibility, due to the additional destaining step in its protocol.
