What are the advantages of CRISPR-Cas9 systems over other genome editing tools?

CRISPR-Cas9 offers superior gene-editing efficiency; CRISPR-Cas9 systems can accurately target and modify specific sequences within the genome, leading to desired genetic changes with a greater success rate compared to other gene-editing techniques. Another advantage is that it exhibits reduced off-target effects. It has been optimized to minimize such off-target effects, resulting in more precise gene editing with fewer unintended changes elsewhere in the genome. A third advantage is that it avoids DNA integration. Unlike some other gene-editing methods, CRISPR-Cas9 does not involve the permanent integration of foreign DNA into the host genome. Additionally, because CRISPR-Cas9 has the ability to cut DNA strands independently, it doesn't require additional cleaving enzymes like other gene-editing tools do. Moreover, CRISPR-Cas9 enables the targeting of multiple genes at once, offering a distinct advantage over other gene-editing techniques. Lastly, CRISPR-Cas9 can be paired with custom-designed guide RNA (gRNA) sequences, directing it to specific DNA targets. A vast library of these gRNA sequences has been generated and is accessible to researchers.
