One advantage of the two-step approach is it offers flexibility by allowing separate optimization of the cDNA synthesis and qPCR steps. This enables researchers to choose different enzymes and reagents for each step, and to control sequence representation. It also helps for optimizing challenging RT-qPCR experiments and for increasing efficiency.
