Primary explant is a technique developed by Ross Harrison in 1907 for the extraction of cells from tissue. This process involves the following steps:
- Step 1: Tissue sample is suspended in basal salt solution.
- Step 2: Carefully slice tissue into pieces and wash by settling.
- Step 3: Remove the basal salt solution.
- Step 4: Transfer tissue pieces on to a suitable growth medium.
- Step 5: Incubate 3 to 5 days and replenish medium as needed.
- Step 6: Separate explants and transfer to a new culture vessel with fresh growth medium.
