How do I conjugate an antibody with phycoerythrin (PE)?

Posted July 9, 2018

I have 100 μg of my antibody supplied in 500 μL of PBS, pH 7.2, and it contains 50% glycerol and 0.02% sodium azide.

Answer

For labeling antibodies with PE, use Buccutite™ Rapid PE Antibody Labeling Kit (Cat# 1310). Our Buccutite™ labeling kits are optimized to label microscale quantites (25 μg or 100 μg) of antibody per reaction.

Before you do your conjugation you will need to perform dialysis on your antibody solution to remove impurities such as glycerol. We recommend keeping the glycerol concentration <10%. The 0.02% of sodium azide is ok.

Based on your antibody specifications, your antibody concentration is 0.2 mg/mL. This is considerably low and will greatly decrease the efficiency of your conjugation reaction. It is preferred to have an antibody concentration > 1 mg/mL when making antibody conjugates. We recommend that you dilute the antibody solution with 5 mL PBS and then use a 10,000 MWCO filter to concentrate the solution to ~100 μL before using Cat# 1310 to label your antibody with PE.
