Perforin-1 (sometimes called Cytolysin, Lymphocyte pore-forming protein or PFP) is a protein with a molecular weight of 61 kDa, expressed in the membrane, cytolytic granule and extracellular region of cells. In humans, perforin-1 is involved in the positive regulation of killing of cells of other organism. Perforin-1 recognizes calcium ion and identical protein, and additionally, takes part in organismal processes, in particular, immunological synapse formation, protein homooligomerization and apoptotic process. Sequencing of perforin-1 has demonstrated it contains 3 types of conserved structural units: EGF-like, C2 and MACPF domain. Perforin-1 has been closely linked to vital functions such as wide pore channel activity. It is clinically significant because abnormalities in its function have been associated with diseases like familial hemophagocytic lymphohistiocytosis 2 (FHL2). Familial hemophagocytic lymphohistiocytosis 2, an autosomal recessive inheritancedisorder characterized by hepatomegaly, hypofibrinogenemia and lymphadenopathy, has in particular been of interest to researchers.
