Anti-Procalcitonin (PCT) antibody *Mouse anti-human, monoclonal IgG1*

Procalcitonin (PCT) is a peptide precursor of calcitonin and is composed of 116 amino acids. It is produced by the C cells of the thyroid and by the neuroendocrine cells of the lung and the intestine. Under normal conditions, the level of PCT in the blood of healthy individuals is very low (< .05 ng/mL) with a half-life of 25 to 30 hours in serum. In response to a pro-inflammatory stimulus, such as a pathogen, the level of PCT may increase as high as 100 μg/mL making it a suitable biomarker of bacterial infection. In clinical biology, PCT has shown to be a useful tool in the diagnosis of sepsis. Anti-PCT monoclonal antibodies are highly specific for the detection of human procalcitonin. They can be used for a broad range of immunoassays such as chemiluminescent immunoassays (CLIA).

Clinical comparison of AAT Bioquest&rsquo;s PCT immunoassays and VIDAS<sup>&reg;</sup> B.R.A.H.M.S PCT assay: 70 clinical blood samples were separately tested using four MAb pairs on AAT Bioquest&rsquo;s CLIA platform and compared to a diagnostic kit from bioMerieux. Data from this study were analyzed using the Passing-Bablok regression method and are summarized in the following scatter plot. Results reveal good agreement between AAT Bioquest&rsquo;s immunoassay and comparison assay. The Mab pairs used are (capture-detection): Mab pair A: Cat# V100120 (Clone 3) - Cat# V100120 (Clone 2); Mab pair B: Cat# V100120 (Clone 4) - Cat# V100120 (Clone 2); Mab pair C: Cat# V100120 (Clone 3) - Cat# V100120 (Clone 5); and Mab pair D: Cat# V100120 (Clone 3) - Cat# V100120 (Clone 6).

Calibration curve for procalcitonin in sandwich chemiluminescence immunoassay (Six calibrator concentration: 0, 0.1, 0.5, 2, 10, 50 ng/mL). All monoclonal antibodies were tested in pairs as capture and detection antibodies to determine the best two-site MAb pairs for the development of quantitative sandwich immunoassays. Detection antibodies were labeled with horseradish peroxidase (HRP).
