The 9 steps of the procedure for the kinyoun-cold method are as follows. First, make a smear of the organism on a slide and let it air dry for 10 minutes. Next, cover the smear with carbolfuchsin for 3 to 5 minutes at room temperature. Lightly rinse the slide with water. For 3 minutes, run 1% sulfuric acid decolorizer over the slide. Then, rinse the slide with water and decolorize it for 1 to 2 minutes until the solution runs clear. Rinse the slide again with water. Cover the smear with the counterstain, methylene blue, for approximately 1 minute. Rinse the slide again with water. After the slide is blotted dry with bibulous paper, it can then be observed under a microscope.
