Endogenous biotin can lead to background staining issues in immunohistochemistry (IHC) staining methods. While formalin fixation and paraffin embedding generally decrease endogenous biotin levels, tissues like the liver and kidney may still retain residual biotin activity. Another drawback is that the LSAB method involves multiple steps, including incubation with streptavidin-biotin complex and subsequent detection steps.
