One disadvantage is that when compared to nitrocellulose and PVDF membranes, nylon membranes tend to have more background noise (particularly with RNA probes). Nylon membranes can also show increased non-specific binding with certain proteins, requiring adjustment of blocking and washing protocols to minimize interference. Additionally, nylon membranes may not offer the best sensitivity for detecting proteins present in low abundance. Nylon membranes also lack selectivity, allowing larger particles to pass through while blocking smaller ones. Consequently, they require regular cleaning after each use to prevent clogging.
