What are the advantages of PEGylated liposomes?

Answer

PEGylated liposomes are liposomes that are modified with polyethylene glycol (PEG). These modified liposomes offer several advantages including: 

  • PEGylation increases the hydrophilicity of the liposome surface and lowers its affinity with phagocytes, allowing it to escape detection by the reticuloendothelial system and avoid being captured. 
  • PEGylated liposomes lower the chances of triggering immune responses, making them useful in delivering macromolecular drugs that could otherwise cause immune reactions.
  • PEGylated liposomes impact the physical and chemical properties of drugs, extending their half-life and reducing liver clearance rate. This allows for lower drug dosage and less frequent administration, while also enhancing the drug’s efficacy and minimizing adverse reactions. 
  • PEGylated liposomes can protect drugs, such as Doxil®, from being absorbed by non-target myocardial cells, lowering the incidence of cardiac toxicity.
  • When used in mRNA vaccines, PEGylated lipids stabilize the lipid bubbles that encapsulate the mRNA.
