How do I choose the right lysis buffer for my western blot experiment?

When selecting a lysis buffer, the main thing to consider is whether the chosen antibody can recognize denatured samples. If the antibody cannot do so, this information will be specified on the antibody datasheet. In such cases, it's best to use buffers that either lack detergent or contain gentle non-ionic detergents like NP-40 or Triton X-100.  Additionally, depending on the protein's location, different lysate buffers may be required to achieve high yield and purity. However, since each protein behaves uniquely, it may not always solubilize effectively in standard buffers. Thus, experimenting with different buffers and adjusting detergent types can help optimize protein solubility.
