Do I have to fix cells before DAPI staining?

Answer
It is preferred, but not required to fix cells before DAPI staining. DAPI is toxic to cells, and thus it is preferred to be used on fixed cells as opposed to live cells. It is a cell permeable dye and it has the capability to enter live cell membranes. It also passes less efficiently in live cell membranes as opposed to fixed cells.
