Why is the Total GSH standard solution concentration equal to twice that of the GSSG standard solution concentration? Why is [GSSG] equal to ([Total GSH] - [GSH]) DIVIDED by 2?

Answer
After reduction, 1 mole of GSSG becomes 2 moles of GSH. Thus, if the initial GSSG concentration is 1 µM, the GSH concentration after reduction is 2 µM. Conversely, to determine [GSSG] from [Total GSH], you have to divide by two to account for the GSSG to GSH molar ratio: 1 GSSG ⟶ 2 GSH.
