In terms of techniques such as immunohistochemistry and immunofluorescence, direct detection involves using a primary antibody that is labeled, while with indirect detection, the secondary antibody is labeled instead. Indirect detection may be preferred if the antigen is poorly expressed and the signal needs to be amplified; however, indirect detection can result in higher cross-reactivity and higher background compared to direct detection.
