A basic immunofluorescence protocol for expression of early endosome and lysosome includes fixation with methanol in -20C for 5 minutes, permeabilization with 0.1% saponin or 0.1% TritonX-100 in PBS for 10 min, and blocking with 1% BSA in PBS for 30 min. For early endosome expression, you can incubate with an anti-Rab5 antibody; for lysosomes, you can incubate with an anti-LAMP1 (lysosomal associated membrane protein-1) antibody. This incubation can be done overnight at 4C.
