Anti-Neutrophil gelatinase-associated lipocalin (NGAL) antibody *Mouse anti-human, monoclonal IgG1*

Neutrophil gelatinase-associated lipocalin (NGAL) is a protein expressed mostly in neutrophils. It is also present in lower levels in the kidney, prostate and epithelia of the respiratory and alimentary tracts. NGAL is a vital component in the innate immune response to bacterial infection. It limits bacterial growth by sequestering iron-containing microorganisms. NGAL's small size and protease resistant properties makes it easily excreted and detected in the urine. Under normal conditions, NGAL levels in the urine and plasma are low. However, in response to kidney injury NGAL levels increase sharply reaching diagnostic levels quickly. In clinical biology, NGAL is a novel biomarker of acute kidney injury (AKI) and is used in the early diagnosis of chronic kidney disease. Anti-NGAL monoclonal antibodies are highly sensitive for the detection of human NGAL. They can be used for a broad range of immunoassays such as chemiluminescent immunoassays (CLIA), latex enhanced turbidimetric immunoassays (LETIA), lateral-flow immunoassays (LFIA) and ELISA.

Calibration curve for human NGAL in latex-enhanced turbidimetric immunoassay: a set of NGAL calibrators were reacted with specific antibodies coated onto microparticles to form insoluble complexes that were measured turbidimetrically at a series of different wavelengths.
Determination of clinical&nbsp;samples using four MAb combinations on CLIA platform. A study was performed where 50 specimens were tested using different antibody combinations (A, B, C and D) on AAT Bioquest&rsquo;s CLIA platform and compared to a high-quality competitor&rsquo;s assay. Results showed the four combinations have correlation coefficients (r) &gt; 0.90. The MAb combinations used are (capture-detection): MAb combination A: Cat# V100000 (Clone 1) &ndash; Cat# V100005 (Clone 1); MAb combination B: Cat# V100005 (Clone 1) - Cat# V100000 (Clone 1); MAb combination C: Cat# V100005 (Clone 2) - Cat# V100000 (Clone 1); and MAb combination D: Cat# V100005 (Clone 3) - Cat# V100000 (Clone 1).
In order to meet the application of AAT Bioquest&rsquo;s anti-NGAL MAbs on LETIA platform, 40 clinical blood samples were separately tested using mixed MAbs on AAT Bioquest&rsquo;s LETIA platform and compared to a competitor&rsquo;s diagnostic kit. The correlation coefficient between the two systems is over 0.90.
